Background
Somatostatin receptor 2 (SSTR2), encoded by the SSTR2 gene on chromosome 17q25.1, is a 369-amino-acid Gi-coupled class A G-protein-coupled receptor (GPCR) with seven transmembrane helices, extracellular N-glycosylation sites and intracellular phosphorylation motifs, existing as two splice variants (SST2A/B) with SST2A predominating in human tissues; it is highly expressed in brain and endocrine organs (pancreatic α/β-cells, pituitary somatotrophs) where it binds somatostatin-14/28 to inhibit adenylyl cyclase, voltage-gated Ca²⁺ channels and hormone secretion (e.g., glucagon, insulin, GH, TSH), and is widely over-expressed in neuroendocrine tumors, making it both a prognostic marker and a therapeutic target for radiolabeled peptides or selective agonists such as octreotide to suppress tumor growth and induce apoptosis .
